In the Book In the Movie
The Vampires have no leader. Adam is the leader of the Vampires.
When Abraham is eleven years old, his father tells him about vampires. Lincoln is told by Henry Sturgess about vampires after he is rescued from one at the age of twenty.
Lincoln at age twelve, kills the vampire who was responsible for his mother's death. The owner of the plantation is Jack Barts, a vampire, who poisoned Lincoln's mother. Abe tries to get revenge at age twenty, but fails. He trains as a hunter for ten years and eventually gets his revenge on Barts and kills him.
Abe sees the slaves as the life blood of the Vampires and becomes an Abolitionist in order to stop the flow of slave blood. Abe wants to end slavery because of his black friend from childhood, Johnson. He is warned by Sturgess to stick to the hunting plan.
Willie Lincoln is killed by a vampire "assassin". He is bitten by Vadoma, Adam's sister as a way to mock Abe.
Edgar Allan Poe is a friend of Abe's and warns him about the Vampires use of slaves. Poe is later murdered by a vampire. Poe doesn't appear.
Lincoln signs the Emancipation Proclamation as a way to starve the Vampires and helps turn the tide of the Civil War. Abe frees the slaves and that makes the Vampires go crazy and start attacking all humans.
A man named Jack Armstrong helps Lincoln hunt vampires for some time. No Jack Armstrong.
Henry assigns Abe three vampire bodyguards. Abe's only companions are Willie and Joshua Speed.
Abe overhears his father and Barts talking about his father's debt and foreshadows his mother's death. Abe sees Barts biting his mother when she's sleeping.
Vampires possess heightened strength and may kill other vampires. The vampires are given additional abilities (invisibility, for instance) and a weakness to silver, causing the climatic scenes to concern the aquisition of enough silver to defeat the vampires.
After John Wilkes Booth, a vampire, assassinates Lincoln, Henry turns Abe into a vampire, saying that some people are just to interesting to die. They fight together through World War II, and the last scene they are attending MLK's "I have a dream" speech. Film ends with Lincoln and Mary Todd leaving to go the Ford's theater. The movie fast forwards to present time where Henry is in a bar and says the same thing when he met Abe.
Abe is a vampire and attends Martin Luther King's "I have a dream" speech. He is assassinated and doesn't become a vampire.
